Homologous recombination (HR)-mediated DNA repair is a prerequisite for the maintenance of genome stability. Cancer cells with HR deficiency (HRD) are selectively and significantly eliminated by poly (ADP-ribose) polymerase inhibitors (PARPi), which benefit the survival rate of cancer patients, including breast, ovarian, pancreatic, and prostate cancers. To date, sequencing the pathogenic mutations in HR-associated BRCA1/2 genes and analyzing the genomic scar derived from genome instability have been used as the clinical biomarker for the treatment of PARPi.
