Title: Tissue-specific transcriptome assembly of sensory organs in the Formosan subterranean termite Coptotermes formosanus

The Formosan subterranean termite, Coptotermes formosanus, is a widespread eusocial insect and one of the most destructive wood-feeding pests of buildings worldwide. Although its complex social behaviors and chemical communication systems have been studied extensively, high-resolution transcriptomic data that link gene expression to specific sensory and non-sensory tissues have remained unavailable. Here we report a comprehensive tissue-specific transcriptome resource generated by Illumina RNA-seq from five distinct organs (antennae, maxillary and labial palps, legs, heads, and remaining body parts) pooled from 600 laboratory-reared worker individuals. The final de novo assembly yielded 184,092 non-redundant unigenes. BUSCO assessment against the Insecta odb10 dataset indicated 98.56% completeness (29.7% single-copy and 68.9% duplicated BUSCOs), with 1.23% fragmented and 0.20% missing. Functional annotation assigned KEGG pathways, conserved domains, and transmembrane structures to approximately 57% of the unigenes. This high-quality dataset provides a valuable foundation for future research on the molecular basis of chemosensation in eusocial insects, the evolution of social signaling, and the development of targeted behavioral or molecular strategies for termite control. All raw reads, assembled unigenes, expression matrices, and annotation files are publicly accessible to support comparative transcriptomics and pest-management studies within the order Blattodea. This dataset include a C. formosanus unigene data (.txt file) and annotated genes(.xls file).
