Brain region-specific changes in neurons and glia and dysregulation of dopamine signaling in Grin2a mutant mice [snRNA-seq]

To identify which brain cell types are contributing to the bulk transcriptomic changes in Grin2a mutant mice, we performed single-nucleus RNAseq (snRNAseq) in Grin2a heterozygous and homozygous mutants and their wild-type littermates at 4 and 12 weeks of age. SnRNAseq analysis of prefrontal cortex (PFC), somatosensory cortex (SSC), hippocampus (HP), striatum (ST) and Thalamus (TH) at 12 weeks, as well as PFC and HP at 4 weeks in Grin2a mutant mice.

为明确参与Grin2a突变小鼠整体转录组变化的脑细胞类型,我们于4周龄与12周龄的Grin2a杂合突变、纯合突变小鼠及其野生型同窝仔鼠中开展了单细胞核RNA测序(single-nucleus RNAseq, snRNAseq)。针对12周龄的Grin2a突变小鼠,我们对其前额叶皮层(prefrontal cortex, PFC)、躯体感觉皮层(somatosensory cortex, SSC)、海马体(hippocampus, HP)、纹状体(striatum, ST)及丘脑(Thalamus, TH)进行了snRNAseq分析;同时对4周龄的该类小鼠的前额叶皮层与海马体开展了相同的测序分析。
