RNAseq was used to identify host and viral transcriptome changes in methionine restricted Rael cells or Rael cells expressing control, AHCY or MAT2A sgRNAs. For methionine restriction, Rael cells were washed with PBS and then cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.01mM L-methionine for 3 days. Control cells were cultured in methionine free RPMI-1640 media supplemented with 10% dialyzed FBS and 0.1mM L-methionine for 3 days. Control or methionine restricted cells were then subjected to RNAseq analysis. For CRISPR KO, Rael cells expressing control, AHCY, or MAT2A sgRNAs were subjected to the RNAseq analysis.
