SOILRECON Project: 16S and 18S Raw Sequences

SoilRecon is a portuguese project funded by FCT national funds that will allow expanding the limits of current knowledge by implementing a balanced approach between global and regional observational data, and predictive models that can improve the way researchers understand the macroecological patterns of soil biodiversity and functions and contribute to the conservation of soil biodiversity and soil ecosystem processes. This dataset corresponds to the sequencing of soil samples from a regional large-scale sampling of soil biodiversity and ecosystem function in the northern region of Portugal. Sampling scheme implemented was a hierarchical stratified system considering first a 20x20km grid across the entire region and second, for each grid cell, randomly selecting six sampling sites, one per major land-use type, i.e., urban areas, pastures, agricultural areas (subdivided into permanent and annual crops), and forests (subdivided into native and non-native forests). Prokaryotic and eukaryotic small subunit ribosomal RNA(SSU rRNA) genes were amplified using primer pairs 799F (5’-AACMGGATTAGATACCCKG-3’) /1193R (ACGTCATCCCCACCTTCC-3’) (Bodenhausen et al. 2013) and Euk_1391f (5’-GTACACACCGCCCGTC-3’)/EukBr (5’-TGATCCTTCTGCAGGTTCACCTAC-3’)(Stoeck et al. 2010) reverse, respectively, and sequenced at the Western Sydney university Next Generation Sequencing facility using the Miseq platform.
