Nonalcoholic fatty liver disease (NAFLD) represents the most prevalent form of liver disease, exacerbated by hypoxia and high-fat diet. This study investigates the role of Angptl4 in Kupffer cells under these conditions. Using a murine NAFLD model and a Kupffer cell line treated with palmitic acid, we analyzed lipid accumulation, inflammation, and oxidative stress. Our study shows that hypoxia and high-fat conditions significantly increased liver triglycerides, hepatic inflammation, and oxidative stress. Angptl4 expression was notably elevated in Kupffer cells, and its silencing reduced lipid accumulation, oxidative stress, and inflammation. These findings suggest that Angptl4 plays a crucial role in NAFLD progression under hypoxic and high fat diet.
