To investigate the potential role of the PTPRZ1 in tumor development, we leveraged CRISPR/Cas9 to perform knockout experiments in two distinct LUSC cell lines, H520 and HCC95, both characterized by high cellular expression of the gene. Following the disruption of PTPRZ1, we observed a significant reduction in the proliferation of both LUSC cell lines, as indicated by the Cell Counting Kit-8 (CCK-8) assay .To comprehensively assess potential off-target effects of the CRISPR/Cas9 editing, we conducted whole-genome sequencing comparing the HCC95-NC and HCC95-KO-PTPRZ1cell lines, which enabled genome-wide evaluation of editing specificity Overall design: WGS of the HCC95-NC and HCC95-KO-PTPRZ1 cells.
