Large-Scale Multiplexed Mosaic CRISPR Perturbation in the Whole Organism

Atlases cataloguing transcriptome landscape across diverse mouse tissues have now been constructed, setting the stage for conquering a new frontier, which is to map "Perturb-Atlases" profiling the effects of genome perturbation on the landscape, thus establishing the genotype-phenotype relationship. To this end, we have developed a transgenic strategy termed iMAP (inducible Mosaic Animal for Purturbation) which enables mosaic CRISPR targeting at the unprecedented, whole-body scale. Here we show thymomas in iMAP-61 mice to investigate the molecular-level effects of genetic perturbations and all of the amplicon data.
