Extract protocol
We isolated cord blood mononuclear cells using density gradient centrifugation with a density of 1.077 g/mL at 266 RCF for 30 min. We then extracted human erythroid cells by CD235-positive magnetic separation.
Library construction protocol
The scRNAseq was performed using the BD Rhapsody™ System mRNA Targeted and Sample Tag Library Preparation Protocol
Data processing
Sample Tag-demultiplexed immune transcriptome data was obtained via BD Rhapsody pipeline ver. 1.10 from R1 and R2 FASTQ files
