Lipids are essential for neuron development and physiology. Lipid droplets (LD) play a central role in coordinating cellular lipid distribution and metabolism in non-neuronal cells; however, there is no general consensus on a role for neuronal LD under normal physiological conditions. We have previously shown that neuronal LD exists in wildtype flies under normal physiological condition, and that the loss of a triglyceride lipase, brummer, increases neutral lipids in the brain (Wat et al., 2020). To investigate the importance of brummer in regulating neuronal metabolism and function in a sex-specific manner, we performed transcriptomic profiling in adult male and female whole brain with RNAi-mediated neuron-specific knock down of brummer. To assess the transcriptomic effects of brummer knock down in either males or females, we raised male and female flies with neuron-specific expression of RNAi targeting brummer transcripts (elav-GAL4>UAS-bmm-RNAi). Adult whole brains were dissected at day 5 to 6 after eclosion for transcriptomic profiling. We also profiled the transcriptome of whole brains from elav-GAL4>+ and +>UAS-bmm-RNAi males and females as controls.
