AlkB homolog 8 (ALKBH8), an enzyme involved in mcm5U modification, catalyzes the methylation of 5-carboxymethyluridine (cm5U) to mcm5U in tRNA. To elucidate the effects on tRNA mediated by Alkbh8 modifications, we conducted an analysis using tRNA-seq focused on E13.5 in Alkbh8-knock out mice. In small RNA extracted from embryos at E13.5, 171 tRNAs were detected; 76 tRNAs increased, and 11 tRNAs decreased in Alkbh8-knock out compared to WT. In Alkbh8-KO mice, the action on cytoplasmic tRNA and the potential to alter the mitochondrial tRNA balance have been suggested.
