Transcriptional analysis of dendritic cells upon TLR stimulation reveals an immunosuppressive profile (RNA-Seq)

Dendritic cells (DCs) simultaneously prime and dampen the immune response. The transcriptional program that regulates immunosuppression in DCs is currently unknown. Here we identified ETS homologous factor as a key transcription factor that regulates DC immunosuppression. Purified splenic DCs (~20%) were plated in complete RPMI-1640 (RPMI-1640 with 10% FCS, 1% L-glutamate, 1% penicillin/streptomycin, and 55 μM 2-mercaptoethanol), and stimulated with 100nM CpG-B at 37 ℃ for 9 hrs. Then, cDCs (Lin- MHC II+ CD11c+) were FACS-sorted and RNA-seq library preparation.
