HIF1A regulated genes in MCF10A cells after treatment with Oncostatin M or control ligands

We employed a single-cell RNA sequencing approach to study the role of Oncostatin M (OSM) in inducing collective cell migration in MCF10A cells and its upregulation of HIF1A. To identify HIF1A-dependent transcriptional programs, we stably knocked down HIF1A using shRNA and treated cells with OSM. Overall design: MCF10A breast epithelial cells stably expressing shHIF1A or control shSCR were grown in vitro and treated with Oncostatin M (OSM), epidermal growth factor (EGF), or a combination of EGF and interferon gamma (IFNG) for 24 hours prior to single-cell RNA sequencing.
