To increase the vitamin C content of lettuce in facility cultivation , UV-B irradiation was used to increase the accumulation of ASA in crops. Three UV-B dose treatments: 0, medium dose 7.2 and high dose 12.96 KJ/m2/d, were selected to perform for transcriptome sequencing, The main conclusions of previous studies are as follows: after the intervention ofthe three methods and three acupoints technique, 1.226 differentially expressedgenes were detected in DRG by RNA-Seq, which mainly enriched on Wnt, IL-17 andMAPK signaling pathways; 2.it could up regulate inhibitory neurotransmitters anddown regulate inflammatory factors; it preliminarily answered the partialmechanism of Tuina in the treatment of neuropathic pain. How Tuina initiates theanalgesic mechanism has become a new scientific issue. This research will selectchronic constriction injury of sciatic nerve (CCI) rats as the model, and utilizea Tuina-technique simulator to simulate three methods ( plucking, point-pressing,and kneading method) on three acupoints (Yinmen, Yanglingquan, Chengshan), andchoose DRG and dorsal horn as the main observation sites. Then, behavioralresearch and morphological observation will be used to looking for thephenotypic evidences; RNA-Seq will be used to detect the differentially expressedgenes at different times and screen the analgesic genes combined with GO functionand KEGG pathway; RT-PCR, Western Blot and immunofluorescence will be used tofurther detect the expression of related genes and their roles in the pathway, soas to clarify the initiation genes and their expression times and sites. Finally,answer the question of "how does Tuina initiate the analgesic mechanism".
