Role of TCHHL1 (trichohyalin-like 1) on the proliferation of normal human epidermal keratinocytes

TCHHL1 (trichohyalin-like 1) protein is a novel member of the fused-type S100 protein gene family. Previous findings suggest that TCHHL1 may play a role in the proliferation of keratinocytes. In the present study, to clarify a role of TCHHL1 in the proliferation of keratinocytes, we performed global-scale gene expression analysis in TCHHL1-Knockdown keratinocytes using an Affymetrix GeneChip® system. Normal human epidermal keratinocytes (negative control siRNA), and normal human epidermal keratinocytes (siRNA for TCHHL1) were used. Total RNA samples were prepared from the cells, and quality of the RNA was analyzed using a Bioanalyzer 2100. Gene expression was analyzed by an Affymetrix GeneChip® system with a Human Genome U133-plus 2.0 array. Sample preparation for array hybridization was carried out as described in the manufacturer’s instructions.
