The goal of this study was to understand how PHRF1 regulates gene expression in HCT116 cells. By conducting RNA sequencing of 3 WT and 3 KO (PHRF1 knockout) samples, we found that DNA damage response pathways were upregulated and splicing-related pathways were downregulated in KO cells versus WT cells. PHRF1 deletion in HCT116 cells was generated through CRISPR-Cas9 mediated gene editing. Total RNA was extracted from 3 WT and 3 KO Hela cell populations and prepared for RNA sequencing. Differential gene expression analysis based on the sequencing data was conducted, comparing gene expression in PHRF1 KO cells to WT cells.
