High-throughput RNA sequencing has been used to comparatively analyze the transcriptome of the MC3T3-E1 cells cultured on cell-adhesive nanofibers and membranes. Compared with membranes, the nanofibers mediate the elongation of laden cells, leading to 905 differentially expressed genes (DEGs). The DEGs related to cytoskeleton, promotion of cell cycle progression, cell adhesion and cell proliferation, are higher expressed in the cells on nanofibers, while DEGs involved in cell-cycle arrest and osteoblast mineralization are upregulated in the cells on membranes. Besides, the expression of most DEGs associated with cell activities is closely correlated with that of cytoskeleton- and cell cycle- as well as cell adhesion-related genes.
