Museomics and targeted sequencing in Orthosiinae (Apocynaceae)

We present a phylogenetic study of Orthosiinae (Apocynaceae) using exclusively herbarium specimens. We optimized DNA extraction protocols and applied rigorous data processing on target capture sequencing (Hyb-Seq) data with an Apocynaceae-specific set of 836 probes. Our results yielded a well-supported species phylogeny comprising approximately 78% of known species and over 50 undescribed specimens. The phylogenetic analysis confirmed the generic relationships and common origin of Orthosiinae species.
