We used scRNA-seq technology to study mouse xenograft tissues from two human TRa1 anaplastic thyroid cancer (ATC) cell lines. Our analysis demonstrated that TRa1 functions as a transcription factor through multiple signaling pathways to suppress tumor growth. Importantly, scRNA-seq analysis showed that TRa1-induced PAX8, via its transcription program, shifts the cell landscape of ATC toward a differentiated state. Overall design: ATC cells isolated from human tumors (11T and 16T) were subcutaneously inoculated into mice to grow into xenograft tumors. Single cells were dissociated from the tumors of the 8-week mice and then analyzed by single-cell RNA sequencing (scRNA-seq).
