The goal of this experiment was to determine whether transplantation of ESC-derived tip-like cells had any discernable impact on gene expression or clustering in non-epithelial cells. Overall design: Adult 129X1/S1 mie were given 1.5U/kg bleomycin intratracheally. 10 days later mice received either 4e6 mCh+ ESC-derived tip-like cells (GFP lentiviral label) or vehicle only (LPM media). At 48 days post-transplantation the lungs of these mice were digested and sorted using FACS. Lungs were dissected down to regions surrounding GFP+ cell transplants (or comparable regions in controls) to enrich for cells potentially impacted by the localized injury and cell transplantation. DRAQ7-/EpCAM-/CD45+ and DRAQ7-/EpCAM-/CD45- cells were mixed at a 1:2 ratio in an attempt to get roughly even mix of immune, endothelial, and other non-epithelial lineages. For each condition, cells were pooled equally from 3 seperate mice in the same cohort.
