Analysis of molecular mechanism of neutrophil differentiation in HL-60 cells: effects of Ak2 hetero knockout, and difference from differentiation into eosinophil

Adenylate kinase 2 (Ak2) is a causative gene of reticular dysgenesis (RD) and, in patients of this disease, Ak2 activity is lost, and neutrophil differentiation is suppressed. HL-60 is a cell line that differentiates into neutorophils when stimulated with ATRA. To elucidate the pathological mechanism of RD, we attempted to generate Ak2 knockout cell lines from HL-60 using CRISPR/Cas9 technology and established two heterozygous knockout cell lines (#5 and #47). To confirm the genomic DNA sequence of those two cell lines, whole genome sequencing analysis was performed. Furthermore, in order to investigate the difference between eosinophil differentiation and neutrophil differentiation, we also performed genome analysis of HL-60 clone 15, which is a subclone of HL-60 cells that differentiate into eosinophil.
