Tumor-associated macrophages (TAMs) play an important role in cancer progression and have been implicated to play a role in prostate cancer. However, TAMs in osteogenic prostate cancer bone metastasis have not been characterized. Here we used a novel osteogenic MycCaP-BMP4 model to study the unique cellular and molecular mechanisms in driving the tumor progression in osteogenic prostate cancer metastasis. MycCaP and MycCaP-BMP4 cells were injected subcutaneiously into FVB mice. Bulk tumors were dissociated into single cells and subjected FACS sorting for CD45+, CD11b+, GR1- and F4/80+ macrophages. Total RNA was isolated from sorted macrophages and subjected to RNA-seq.
