Tgif1 regulates quiescence and self-renewal signaling pathways of hematopoietic stem cells (HSCs)

In this study we demonstrate that Tgif1 has a role in HSCs maintenance, self-renewal and quiescence. RNA sequencing data of LSK cells (HSCs enriched cell population) from Tgif1-/- and wild type mice implicates that multiple pathways involved in HSC quiescence and self-renewal are disturbed in Tgif1 deficient mice. RNA expression profiles of wild type (WT) and Tgif1-/- LSK cells were generated by RNA sequencing, in triplicate, using Illumina HiSeq 2000.
本研究证实,Tgif1在造血干细胞(Hematopoietic Stem Cells, HSCs)的维持、自我更新与静息状态调控中发挥重要作用。对来自Tgif1基因敲除(Tgif1-/-)与野生型小鼠的LSK细胞(富集造血干细胞的细胞群)的RNA测序数据表明,参与造血干细胞静息状态维持与自我更新的多条信号通路在Tgif1缺陷小鼠中发生紊乱。本研究采用Illumina HiSeq 2000测序平台,对野生型(WT)与Tgif1基因敲除LSK细胞的RNA表达谱进行了三次生物学重复的RNA测序。
