The effect of pinto bean supplementation on gut microbiota and metabolic phenotype was assessed. Four-week old, male C57BL/6N mice were obtained from Charles River Laboratory. After two-week acclimation, mice were randomly assigned in a 2 x 2 factorialdesign to one of four dietary treatment groups (n=6 mice/group): Control (C, 10% fat, 10% sucrose, AIN-93G purified diet), C + 10% Pinto Bean (C+PB), High Fat High Sucrose (HFS, 45% fat, 20% sucrose), and HFS + 10% PB (HFS+PB) for 30 days. Food and deionized water were provided ad libitum. Food intake was recorded daily while body weight was monitored weekly. At the end of the study (30 days), mice were fasted for 3 hours, anesthetized with ketamine/xylazine cocktail, cecal contents were collected for microbiota profiling.
