We report differential gene expression in hematopoietic cells (lineage-Sca+) upon overexpression and transplantation of murine hematopoietic stem cells. Overall design: MicroRNA-125a was overexpressed in murine hematopoietic stem cells (LSK/SLAM) and transplanted into lethally irradiated reciepients. Twelve weeks after transplantation, 2000 (lineage-Sca+) bone marrow cells were directly sorted into lysis buffer and snap frozen immediately before moving to library prep.
