Mechanoregulation of the TGFb signaling pathway in response to applied mechanical loads is required for bone anabolism. While fluid shear stress (FSS) is known to be a critical physical cue experienced by osteocytes following bone loading that has been shown to regulate TGFb signaling in other cell types, the extent to which activation of this pathway affects FSS-mediated changes in gene expression remains unknown. In this study, OCY454 cells grown in PDMS microfluidic chambers were pretreated with the TGFb receptor type-I inhibitor 10 uM SB-431542 or vehicle (1% DMSO) for 60 minutes before stimulation with 1 Pa steady FSS for 120 minutes. Control (non-flow) cells were grown in identical conditions but not stimulated with FSS.
