The microcosmic and specific changes of human photoaging skin without chronologic aging is still unclear and rare-ly analyzed. We aimed to assess biological processes of the cell-subgroups alteration and mechanisms of skin pho-toaging using single cell sequencing and biological analysis were used between sun-exposed forearm skin and un-exposed buttock skin in same individual via skin punch biopsy. Forearm skin and buttock skin were obtained from the same healthy individual via skin punch biopsy. After enzymatically and mechanically disrupting the tissue and stringent cell filtration, dead cells were thoroughly removed. Single cell RNA sequencing (scRNA-seq) was used to test the cell populations and subsets. Seurat and other packages in R version was used for data analysis.
