To formally assess how 5mC shapes the H3K27me3 landscape, we profiled the epigenome of naive and differentiated cells in the presence and absence of the DNA methylation machinery. Whole genome bisulphite sequencing and H3K27me3 and H3K36me3 CUT&Tag was performed on mouse embryonic stem cells and epiblast-like cells. H3K27me3 CUT&Run was performed on epiblast-like cells treated by TET1 site-directed epigenome editing.
