The process of regeneration by in vivo transdifferentiation in mammals is poorly understood. Here, using pancreatic ß cell regeneration as a paradigm, we performed a single-cell transcriptomic study of in vivo transdifferentiation from adult mouse acinar cells to induced ß cells. Overall design: Adenovirus (Pdx1, Mafa, Ngn3 co-expression vector, Ad-M3C) infected pancreatic cells are labeled by red fluorescent protein mCherry, and mCherry+ Calcein Blue, AM+ cells are sorted by FACS for scRNA-seq library construction. Acinar cells labeled with Calcein Blue, AM from no treat mice are also sorted by FACS to construct scRNA-seq library for control.
