RNA-seq of non-tumorous liver tissues in LiK and LiKM mice after normal diet or ACR diet

KrasG12D mutation and Mdm2 loss in the liver (LiKM) accelerated liver carcinogenesis in mice, compared with LiK (KrasG12D mutation in the liver) mice. Acyclic retinoid (ACR) diet suppressed tumor development in LiKM mice. The goal of RNA-seq of non-tumorous liver tissues is to identifiy the effect of ACR diet on the transcriptomic profile of the liver and clarify the mechamism of ACR-mediated tumor suppression in LiKM mice. LiK and LiKM mice were fed a normal diet or a diet containing 0.06% ACR from 5 weeks of age until sacrifice at 4 months of age. Non-tumorous liver tissues were obtained at the time of sacrifice.
与仅在肝脏中携带KrasG12D突变的LiK小鼠相比,肝脏同时存在KrasG12D突变与Mdm2缺失的LiKM小鼠可显著加速小鼠肝脏的癌变进程。无环视黄醇(acyclic retinoid, ACR)饮食可抑制LiKM小鼠的肿瘤发生发展。本研究针对非肿瘤性肝组织开展RNA测序(RNA-seq),旨在明确ACR饮食对肝脏转录组谱的调控作用,并阐明ACR介导LiKM小鼠肿瘤抑制的分子机制。实验中将LiK与LiKM小鼠分为两组,自5周龄起分别饲喂正常饲料或添加0.06% ACR的饲料,直至小鼠4月龄时处死。实验于小鼠处死后采集其非肿瘤性肝组织样本。
