UHRF1 is a mediator of KRAS driven oncogenesis in lung adenocarcinoma [CRISPR_screen]

We performed a focused CRISPR in KRAS mutant lung cancer cells (A549) using a minipool sgRNA library targeting 80 lung cancer specific tumor suppressor genes and 12 control genes. This screen identified tumor suppressor genes regulated by Uhrf1 in KRAS mutant lung cancer. Overall design: KRAS mutant A549 cells expressing Cas9 were infected with the minipool sgRNA library containing 483 sgRNAs targeting 92 protein coding genes and 26 negative "safe" sgRNAs. Following puromycin selection the cells were transfected either with a negative sgRNA or sgRNA targeting UHRF1. Cells were expanded for 4-5 days and then split. At this point the cells were also collected for T0. After 18 days of passaging cells were collected for DNA extraction (T1).
