We examined differential gene expression profiles between MCF7 human breast cancer cells treated with vehicle alone (control), palbociclib monotherapy, palbociclib alternating with ICI182,780 treatment, and palbociclib resistant cells. For this, we used gene microarray expression profiling to identify significantly changed genes under different treatment conditions. Gene expression signatures for four conditions of MCF7 cells were: (1) parental cells; vehicle treated (0.04% DMSO); control, (2) cells treated with palbociclib monotherapy at 750nM for 10 weeks, (3) alternating treatment with palbociclib at 750nM for 1 weeek and then switched to ICI182,780 at 750nM for 1 week, for 10 weeks total, and (4) cells cultured for >6 months in palbociclib (500nM). Four biological replicated were collected for each treatment condition.
