MDT-15/MED15 is a subunit of the Mediator complex and is known to regulate specific gene programs. We had previously profiled gene expression in C. elegans following mdt-15 depletion by RNAi. To complement this, we also performed gene expression profiling of the mdt-15(tm2182) hypomorhic mutant to identify MDT-15 regulated genes and developmental and/or physiological gene programs. We grew developmentally synchronized wild-type and mutant worms to the L4 larval stage and harvested them for RNA extraction. L4 was chosen to avoid contamination from embryonic mRNA present in adult worms and to allow best comparison to previous Mediator subunit expression profiling done at the same stage. Please note that this study is an extension of the study profiled in GSE68520; animals were grown, harvested, and processed at the same time. RNA was extracted, purified and processed at the same time, and microarrays were run and analyzed at the same time. The set of wild-type samples for these one-color arrays is identical to the set used in GSE68520.
