Resistance to anti-EGFR therapies poses a significant challenge in the management of metastatic colorectal cancer (mCRC) patients. Our previous research has illustrated that AURKA phosphorylates YAP1, leading to increased stability and the induction of stemness, consequently promoting resistance to cetuximab. The AURKA inhibitor alisertib has demonstrated its ability to reinstate cetuximab efficacy in both cell lines and tumors exhibiting YAP1 activation. Thus, we aimed to investigate the transcriptomic alterations provoked by AURKA inhibition in resistant cell lines to elucidate the underlying changes driving these phenotypic shifts. . The C10 CRC cell line was subjected to treatment with 300nM alisertib for 24 hours. RNA extraction was performed using the NucleoSpin RNA mini kit (Macherey Nagel), followed by the generation of sequencing libraries on the Illumina NovaSeq 6000 platform (Novogene)
