Our research hypothesis was that patient-derived rectal cancer organoids would exhibit dose-dependent radiosensitivity to X-ray irradiation (0–16 Gy) and that image-based analysis would more accurately reflect this sensitivity than the CCK8 assay. The dataset includes two key files: radiation survival.pzf (image-based survival counts of organoids) and radiation cck8.pzf (CCK8 assay absorbance values measured via microplate reader, with 3 biological replicates per dose/time point). The data shows organoids displayed tumor-like heterogeneity and a clear dose-dependent decrease in survival (e.g., 16 Gy reduced survival to ~15% of controls via imaging), while the CCK8 assay failed to accurately capture this response; a notable finding is that image-based analysis generated representative survival curves (with a linear slope, shoulder region, and subsequent straightening), confirming its reliability for assessing organoid radiosensitivity. This data can be interpreted as validating rectal cancer organoids as a viable model for radiosensitivity research and image-based analysis as the optimal assessment method.
