README<br>*** NETosis by Flow Cytometry (Neutrophils) ***<br>To observe whether NETosis is different between our different groups of neutrophils, we used SYTOX green to stain extracellular DNA. We performed NETosis assay on both basal condition and after priming of neutrophils with TNFA. DMSO is used as a vehicle control, and PMA is used to induce NETosis. Due to the high variation between sample, technical quadruplets wereran for each biological sample. Neutrophil samples were analyzed using the MacsQuant10, and gating were performed using FlowLogic version 7. A sample sheet is included in the folder to be able to distinguish the different groups.<br>Note: AgingCohort31 is separated into two folders (AgingCohort21_NTPH_Netosis, AgingCohort31_NTPH_NetosisP2). This batch of samples was processed and ran on together on the same day. However, a MacsQuant10 error caused the instrument to restart, separating the run into two different folders. <br>
