PROBLEM TO BE SOLVED: To provide compositions and methods for modifying and/or regulating the expression of the FAD2 gene in plants， comprising the production of plants in which a desired transgene is targeted and inserted， in the FAD2 (fatty acid desaturase 2) locus involving in fatty acid biosynthesis. SOLUTION: Provided is a method for modifying the genome of a plant cell， the method comprising: cleaving， in a site specific manner， through the use of nucleases binding to any target site of a particular nucleotide sequence in the FAD2 gene in a plant cell， thereby generating a cleavage site in the FAD2 gene， and the FAD2 gene being modified by incorporating an exogenous nucleic acid sequence into the cleavage site after the cleavage.
