To test if HIF pathway is regulated by lipids in zebrafish larvae, we have employed whole genome microarray expression profiling as a discovery platform to identify induced genes upon lalistat or U18666A treatment. Lalistat is a specific inhibitor for lysosomal acid lipase (LAL), which is required for break down triglyceride or cholesterol ester to release free fatty acids. U18666A is a specific inhibitor for NPC, which is required for export free cholesterol oout of lysosomes. Overall design: Zebrafish larvae (5dpf) were treated with vehicle control DMSO, lalistat (25uM) or U18666A(2uM) for 24hr. Each RNA sample were extracted from pooled 4 larvae. Three biological replicates (3 clutches from different pairs of WT zebrafish mating) were conducted.
