Gene expression in Tc1 vs Tc9 cell at day 5 polarization

T cells from spleen and lymphnode were isolated with EasySep Mouse CD8+ T Cell Isolation kit. Isolated CD8+ T cells were stimulated and cultured with plate-bound anti-CD3 and soluble anti CD28 antiboies in Tc1 or Tc9 cell polarizing conditios. After 3-day of polarization, cell were transferred into a new well and culture for another 2-day. on day 5 after seeding, Tc1 and Tc9 cells were collected and sequenced. We used microarrays to detail the global programme of gene expression of Tc1 and Tc9 cells at day 5 polarization. In vitro polarized Tc1 and Tc9 cells were colletected at day 5 for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain genes selected highly expressed in Tc9 cells as cmpared with Tc1 cells.
