Transcriptional responses to IFN-alpha in human dermal fibroblasts

We sought to examine the transcription response to IFNa in fibroblasts from three healthy controls (C1-C3) and a patient with a mutation in STAT2 (P1) Overall design: Cells in 6 well plates were mock stimulated, or stimulated with IFNa (Intron A, 1000IU/mL) for 12h, then washed and rested for 36h, or stimulated for 6h prior to lysis and RNA isolation. Experiments were conducted in biological triplicate.
