DNA from the biomass formed at the oil-water interface from laboratorial experiment. The purity of the extracted DNA was checked using a Nanodrop ND-1000 spectrophotometer (NanodropThermoScientific, Wilmington, DE, USA) (260/280 nmratio), and quantified by a Qubit® 2.0 fluorometer, using the dsDNA BR Assay kit (Invitrogen™), according to the manual. The DNA integrity was also confirmed by electrophoresison 0.1% agarose gel with 1 X TAE buffer. Microbial diversity was analyzed by monitoring composition, abundance and diversity of the bacterial community under different biodegradation treatments of B5, B10, B50 e B100 fuels as received from the distributor and Bushnell-Hass Mineral Medium microcosms for 40 days by Illumina high-throughput sequencing.The bacterial and archaeal V4 regions of 16S rRNA, and fungal 18S rRNA genes were amplified using the universal prokaryotic and eukaryotic primers, respectively, with the addition of a barcoded sequence and the required Illumina adapters.
