Transcriptomic profiling by high-throughput sequencing of primary human Beta cells following SIX2 or SIX3 loss of function. Overall design: Profiling FACS-purified human pancreatic SIX2 or SIX3 shRNA mediated knockdown GFP+ adult Beta cells by RNA-Seq, compared to a non-targeting shRNA control/donor. Profiling FACS-purified human pancreatic SIX3 overexpressing Flag+ juvenile Beta cells by RNA-Seq, compared to an empty overexpressing construct.
