Evaluating DNA extraction protocols for bacterial community profiling in waterlogged archaeological wood

The performance of three commercially available DNA extraction kits — Bead-Beat Micro AX Gravity, DNeasy PowerSoil Pro, and MagAttract PowerSoil Pro DNA — was evaluated for their ability to recover high-quality bacterial DNA from waterlogged archaeological wood preserved under anoxic conditions at Guldborg Sund, Denmark. Extracted DNA was assessed in terms of yield and how extraction method influenced downstream analyses, including amplicon sequencing and bacterial community composition.
