This dataset originates from two ethically approved <b>in vivo</b> studies conducted in male Lewis rats using the adjuvant arthritis (AA) model. All procedures complied with <b>EU Directive 2010/63/EU</b>, the <b>European Convention for the Protection of Vertebrate Animals</b>, and institutional <b>3Rs</b> requirements. Arthritis was induced via intradermal injection of <i>Mycobacterium butyricum</i>. Animals were then treated orally with <i>Citrus latifolia</i> essential oil (20 or 40 mg/kg), D-limonene (25 or 100 mg/kg), sub-therapeutic methotrexate (0.3 mg/kg), or corresponding combination regimens.Primary outcomes included biometric parameters, inflammatory cytokines (IL-17A, IL-1β, MMP-9), oxidative-stress markers (CAT, SOD, TEAC, 4-HNE), and tissue GGT activity, measured using validated plethysmometry, ELISA, and spectrophotometric assays. The volatile constituents of <i>Citrus latifolia</i> essential oil were characterised by <b>GC-MS</b>.<b>The dataset from two in vivo studies performed in adjuvant arthritis rats. This study investigated the anti-arthritic and anti-inflammatory potential of Citrus latifolia (CL) essential oil and its principal component, D-limonene (DL), administered alone and in combination with MTX in the adjuvant arthritis (AA) model in Lewis rats. Moreover, the file contains the volatile components identified in Citrus latifolia essential oil, analysed by GC-MS.</b>
