We used Affymetrix microarrays to understand the genome wide differences in gene expression between WT and Ifitm-/- CD4 T cells Overall design: Fresh single splenocyte suspensions were re-suspended in Ammonium Chloride solution (Stem Cell Technologies Inc, US) and CD4+ T cells were separated using negative selection EasySep system (Stem Cell Technologies Inc, US) according to the manufacturer’s protocol. Microarrays were performed on the purified naïve (resting) CD4 T cells from Ifitm-/- and WT controls. These arrays were done twice with independent biological replicates.
