DNA methylation analysis of breast cancer patient-derived xenografts and organoids

To understand if the generation of xenograft and organoid models of breast cancer alters DNA methylation, we compared the genome-wide methylation profile of matching patient tumors, patient derived xenografts, and organoid cultures derived from xenografts. Reduced Representation Bisulfite Sequencing (RRBS) or Reduced Representation Enzymatic Methylation Sequencing (RREMS) was performed on DNA extracted from 11 sets of matching patient tumor samples, patient derived xenografts, and organoid culture derived from the xenograft tumors.
