This study investigates the role of cGAS signaling in amyotrophic lateral sclerosis using human iPSC-derived microglia-motor neuron co-cultures and TDP-43 Q331K mouse models. Bulk RNA sequencing and single-nucleus RNA sequencing were performed to assess transcriptional and RNA splicing changes associated with TDP-43 pathology and their modulation by pharmacological cGAS inhibition.
