This sequencing project was intended to quantify gene expression in recombinant inbred lines (RILs) of the copepod Tigriopus californicus. RILs were generated by crossing individuals from two populations from Southern Califonia, San Diego (SD) and Bird Rock (BR) in a reciprocal design. After the second generation was produced, individual clutches of eggs (which are always composed of full siblings) were isolated to initiate lines. The lines were then propagated in parallel with strict full-sib mating per generation, for a total of 5 generations of full inbreeding. After the fifth generation, each line was allowed to mate freely to increase the number of individuals, generating inbred RILs that can be used for genotype-phenotype association studies. Once lines had sufficient number of copepods, heat stress (33C) and control (20C) were performed in pools of 20-30 adults (equal sex ratios) and total RNA isolated for RNA-seq libraries.These inbred lines are the same as the ones used for whole-genome sequencing in BioProject PRJNA1054307.
