The intestine is the largest digestive and immune organ in the body and is rich in microorganisms.The gut microbiota is an integral part of the human body that has evolved along with its host. There is growing evidence that although part of the microbiota is conserved, the dynamic changes occur in intestine at different stages and health conditions.The gut microbiota promotes the development of metabolic system and gut immune system . The gut dysbiosis can lead to a variety of diseases such as renal diseases, obesity , type 2 diabetes mellitus hypertension , necrotising enterocolitis of the small intestine (NEC) and inflammatory bowel disease (IBD).Gut microbiota and its fecal metabolites could be affected by medications. Trametes robiniophila murr, also known as Huaier, is a medicinal fungus of which the main active ingredient is Huaier polysaccharide(HR). It was reported that polysaccharides have the properties of immunomodulation in vivo and in vitro. They are considered to be safe immunomodulators. It can activate a variety of immune cells to improve their anticancer effects and also inhibit excessive autoimmune responses. Huaier polysaccharides has the effects of significantly enhancing immunity, inhibiting tumor cell proliferation and metastasis, promoting tumor cell apoptosis and reversing drug resistance.Huaier polysaccharides can play a role at multiple levels by increasing the CD4+/CD8+ ratio, reversing the Th1/Th2 ratio, increasing the number of NK cells, enhancing phagocytosis of macrophages, and regulating the expression of immune molecules. However, the specific mechanism of immune regulation by Sophora japonica has not been elucidated, and whether it affects the gut microbiota is still unknown. The aim of this study was to investigate the effects of Trametes robiniophila murr on gut microbiota and its metabolites.
