Neurotransmitters are increasingly recognized to play important roles in limiting anti-tumor immunity. To verify the biological function of N-acetyl-aspartyl-glutamate (NAAG) in breast cancer, we treated breast cancer cell line SUM159 cells and bone marrow cells with 100uM NAAG for several days. We demonstrated that NAAG significantly promoted breast cancer cell migration and invasion abilities, and disrupted anti-tumor immunity by rewiring the myeloid progenitor differentiation of polymorphonuclear myeloid-derived suppressor cells, which jointly promoted breast cancer growth and metastasis. RNA-seq analysis results displayed that MAPK and Notch signaling were activated by NAAG treatment, which were further verified at the protein levels.
